研究内容

Within the maize kernel, the endosperm takes up and stores nutrients from the mother plant, and additionally, serves to nourish the growing embryo via nutrient transfer into the scutellum (cotyledon). In my PhD research, I asked how nutrient reallocation from the mother plant to the endosperm and embryo is regulated.

We cloned the gene underlying the classical endosperm-filling defective mutant floury3, and showed this encodes a PLATZ transcription factor expressed specifically in starchy endosperm cells. We demonstrated that FL3 interacts with RNA polymerase III to regulate tRNA and 5S-rRNA accumulation, and hence, modulates starch and storage protein synthesis (Li, Wang, Ye, et al, 2017). We further showed that Fl3 is paternally imprinted under proximal and distant cis-regulation (unpublished), implying maternal control on nutrient allocation to the seed.

Additionally, we developed an RNAi-based system to alter intra-kernel nutrient reallocation, and showed that down-regulation of α-zeins, the predominant endosperm storage proteins, greatly enhanced the transport of nutrients into the embryo, resulting in an enlarged scutellum. These responses we showed are mediated by the embryo-specific transcription factor VIVIPAROUS1, which thus provides a key link coordinating endosperm nutritional status and embryo growth (Zheng, Li, et al, 2017).

My long-term goal is to establish an independent research career studying embryogenesis in the grasses. To this end, I started my postdoctoral research in the lab of Prof. Dr. Marja Timmermans at University of Tübingen to investigate the role of small RNA signals in the establishment of the shoot stem cell niche during maize embryonic development.
